So If you want to dig deeper into your genetic markers and start to put together your polymorphisms and snp’s then I  purchase the test for 23andme to get started.  Once you take your 23andme test (saliva test and return sample in the mail!) then once you get the results back on their website — you can download your RAW DATA and save to your computer.  Next, I would start uploading your raw data into other website genetic program as DNAFit, Strategene, Genetic Genie and Genopalate.

    2. Enzyme involved in breaking down neurotransmitters as serotonin, norepinephrine and dopamine
    3. When combined with COMT V158M mutation- as below- may result in more severe NT imbalances (would it be related to  Chronic Fatigue?)
    4. Methylation problems often have trouble breaking down NT
  2. VDR Taq
    1. Allele: AA
    2. Vitamin D Receptor for -nuclear hormone receptor for VD3
    3. Chronic illness – show low normal VD3, neurological and immune conditions
    4. Vitamin D stimulates enzymes that create dopamine
    5. May have trouble tolerating methyl donors with COMT V158M
    6. The combo of COMT and VDR can lead to wide range of dopamine levels
  3. SOD2
    1. Allele for me: GG

Genetic Genie Yellow Result:

  1. COMT V158M
    1. Allele: AG
    2. COMT helps breakdown certain NT and catecholamines
    3. Dopamine, epinephrine and norepinephrine
    4. Metabolizing estrogens
    5. Trouble breaking down these NT chemicals from impaired function of the enzyme
    6. May have trouble with methyl donors
  2. COMT H672
    1. Allele: CT
    2. As above- enzyme involved in breaking down important NT
  3. ACAT1-02
    1. Allele: AG
    2. Acetyl co-enzyme A acetyltransferase plays a role in lipid metabolism and energy generation
    3. Depletes B12
  4. MTHFR C677T
    1. Allele: AG
    2. Methylenetetrahydrofolate reductase- helps convert homocysteine to methionine
    3. Mutation in this enzyme may converting homocysteine = high levels
    4. Impaired function of this enzyme can contribute to health conditions (Dr. Lynch)
    5. May lead to high levels of homocysteine – reduced activity of this enzyme if mutations
  5. MTRR A664A
    1. Allele: AG
    2. Methionine synthase reductase helps recycle B12
    3. Combination of MTR and MTRR mutations can deplete methyl B12
    4. MTR provides instructions for making the enzyme methionine synthase
    5. Methionine synthase helps convert the amino acid homocysteine to methionine
    6. Methionine synthase requires B12 (methyl cobalamin form)
    7. MTR mutation increases the activity of MTR gene – resulting in greater need for B12
    8. MTR enzyme causes B12 to deplete since it is using it up at faster rate
    9. MTR mutations – may be underlying cause of methylcarbylamine deficiency
    10. Megaloblastic anemia may occur as consequence of reduce methionine synthase activity
    11. MTHFR and MTR combo = high homocysteine levels unless both treated with B12 and folate
  6. BHMT-02
    1. Allele: CT
    2. Betaine homocysteine methyltransferase acts as shortcut through the methylation cycle to help convert homocysteine to methionine
    3. The activity of this enzyme can be negatively influenced by STRESS
    4. This enzyme is related to methylation
  7. BHMT-08
    1. Allele: CT
  8. CBS A360A
    1. Allele: AG
    2. Cystathionin beta synthase catalyzes the first step in the transculturation pathway from homocysteine to cystathionine
    3. Defects in CBS -upregulation of the CBS enzyme – the enzyme works too fast
    4. Often see low levels of cystathionine and homocysteine due to rapid conversion to taurine- leads to high levels of taurine and ammonia
    5. Shown to result in sulfur intolerance in some patients
    6. BH4 (helps regulate NT & mood) can become depleted with CBS upregulation
    7. Lack of BH4 can lead to mast cell degranulation and possible MCAD
